Overview

Magrolimab is a humanized monoclonal antibody that targets the cell surface antigen CD47. CD47 acts as a "don't eat me" signal on cancer cells by inhibiting phagocytosis by macrophages. By binding to and blocking CD47, magrolimab promotes macrophage-mediated phagocytosis of tumor cells and stimulates both innate and adaptive immune responses against cancer. It has been investigated primarily for hematologic malignancies such as myelodysplastic syndromes (MDS) and acute myeloid leukemia (AML), often in combination with azacitidine. Magrolimab was originally developed by Forty Seven (a Stanford University spin-out) and later acquired by Gilead Sciences[1][2][3][4][6]. As of early 2024, development for hematologic malignancies was discontinued due to lack of efficacy and increased risk of death in clinical trials[8][9].
