Overview

Magrolimab + rituximab is a combination of two monoclonal antibodies: **magrolimab** (a humanized IgG4 antibody targeting CD47) and **rituximab** (a chimeric IgG1 antibody targeting CD20). Magrolimab binds to CD47 on tumor cells, blocking its interaction with signal regulatory protein alpha (SIRPα) on phagocytic cells, thus removing CD47’s “don’t-eat-me” signal and enabling macrophage-mediated tumor cell phagocytosis. Rituximab binds to CD20 on B lymphocytes, mediating direct cytotoxic effects and antibody-dependent immune mechanisms. The combination is being clinically evaluated for its potential synergy, particularly in relapsed/refractory B-cell non-Hodgkin lymphoma (including indolent lymphomas and diffuse large B-cell lymphoma), based on preclinical models and early-phase trials demonstrating antitumor activity and manageable safety.[2][3][7]
