This agent is a **combination of three drugs**: magrolimab (an anti-CD47 monoclonal antibody), rituximab (an anti-CD20 monoclonal antibody), and acalabrutinib (a selective Bruton tyrosine kinase inhibitor). **Magrolimab** blocks the "don't eat me" signal on cancer cells by inhibiting CD47, thereby promoting phagocytosis of tumor cells. **Rituximab** targets CD20 on B lymphocytes, leading to immune-mediated cell lysis. **Acalabrutinib** selectively inhibits Bruton tyrosine kinase (BTK), blocking B-cell receptor signaling essential for B-cell proliferation and survival. Each agent individually shows activity against B-cell lymphomas, and combinations of two of these agents (e.g., magrolimab + rituximab, acalabrutinib + rituximab) have demonstrated efficacy and safety in clinical trials for relapsed/refractory indolent non-Hodgkin lymphoma and follicular lymphoma[1][2][3][4][5][7][8]. There is, however, no published evidence describing the clinical use or trial data for the **triple combination** of magrolimab + rituximab + acalabrutinib as of the current date.
