**Magrolimab + nivolumab** is an investigational combination therapy consisting of *magrolimab*, a first-in-class humanized monoclonal antibody targeting CD47, and *nivolumab*, a monoclonal antibody checkpoint inhibitor targeting the programmed death-1 (PD-1) receptor. - **Magrolimab’s mechanism:** Blocks CD47, a “don’t eat me” signal expressed on cancer cells, thereby enabling macrophage-mediated phagocytosis of tumor cells. - **Nivolumab’s mechanism:** Inhibits PD-1 on T cells, preventing interactions with its ligands (PD-L1/PD-L2) and releasing immune inhibition, facilitating T-cell mediated tumor destruction[2][5]. - This combination leverages both innate (macrophage engagement) and adaptive (T cell activation) immune responses for enhanced anticancer activity. - Developed and under investigation primarily for hematological malignancies and solid tumors, including myelodysplastic syndromes and solid tumors like non-small cell lung cancer. - Both are administered intravenously and studied as part of immuno-oncology regimens.
