MAIT engagers are a novel class of bispecific, tetravalent immune cell engagers developed by Biomunex Pharmaceuticals using their proprietary BiXAb platform. These agents are designed to specifically target and activate Mucosal Associated Invariant T cells (MAITs) by binding to their unique T cell receptor (TCR) and tethering them to tumor-associated antigens on cancer cells. Unlike traditional CD3-based T-cell engagers, MAIT engagers do not activate regulatory T cells (Tregs) or cause broad CD4 T-cell activation, potentially reducing the risk of cytokine release syndrome (CRS) and immune suppression within the tumor microenvironment. Preclinical data demonstrate potency comparable to CD3 engagers with a superior safety profile and efficacy in solid tumor models, including ovarian cancer.
