Overview

MAK2124 is an orally bioavailable small molecule inhibitor developed by MAK Scientific that targets the endocannabinoid system. It specifically inhibits N-acylethanolamine-hydrolyzing amidase (NAAA), an enzyme responsible for the degradation of palmitoylethanolamide (PEA). PEA is an endogenous lipid mediator known for its potent anti-inflammatory and analgesic properties, acting primarily through the PPAR-alpha receptor. By inhibiting NAAA, MAK2124 increases local concentrations of PEA, thereby providing a therapeutic approach for the management of chronic inflammatory and neuropathic pain. The compound is currently in the preclinical stage of development.
