Malformin A1 is a marine-derived cyclic pentapeptide isolated from the fungus *Aspergillus niger*. It has demonstrated potent cytotoxic activity against ovarian cancer, particularly in cisplatin-resistant cell lines. The compound appears to exert its effects by modulating key oncogenic signaling pathways, including the MEK/ERK and NF-κB pathways. Research indicates that malformin A1 can resensitize resistant ovarian cancer cells to apoptosis, potentially overcoming chemoresistance in late-stage gynecological malignancies. Preclinical studies have shown that treatment with malformin A1 results in a significant reduction of total MEK1/2, ERK1/2, and NF-κB protein levels in sensitive cells, while modulating these pathways in resistant cells to trigger cytotoxic mechanisms.
