Overview

MALP-2S is a synthetic derivative of the macrophage-activating lipopeptide-2 (MALP-2), a 2-kDa lipopeptide originally isolated from the cell membrane of *Mycoplasma fermentans*. It functions as a potent and selective agonist of the Toll-like receptor 2/6 (TLR2/TLR6) heterodimer. By binding to these receptors, MALP-2S activates the MyD88-dependent signaling pathway, leading to the activation of NF-kappaB and the subsequent release of pro-inflammatory cytokines and chemokines from macrophages, monocytes, and dendritic cells. Developed primarily by the Helmholtz Centre for Infection Research (HZI), MALP-2S has been investigated for two main therapeutic applications: as a topical agent to accelerate the healing of chronic or infected wounds by stimulating local innate immunity and tissue repair, and as a mucosal vaccine adjuvant to enhance both systemic and mucosal immune responses against various pathogens.
