The MALT1 degrader being developed by Eilean Therapeutics is a small molecule therapeutic designed to induce the targeted degradation of the Mucosa-associated lymphoid tissue lymphoma translocation protein 1 (MALT1). MALT1 is a unique paracaspase that functions as both a protease and a critical scaffolding protein within the CBM (CARMA1-BCL10-MALT1) complex. This complex is essential for mediating NF-κB signaling following antigen receptor activation in B and T cells. While traditional MALT1 inhibitors only address the protein's proteolytic activity, degraders eliminate the entire protein, thereby disrupting both its enzymatic and scaffolding roles. This comprehensive inhibition is intended to provide enhanced therapeutic efficacy in treating B-cell malignancies, such as activated B-cell-like diffuse large B-cell lymphoma (ABC-DLBCL), and various autoimmune or inflammatory conditions driven by dysregulated NF-κB signaling.
