MANA-312 is an allogeneic donor-derived cell therapy developed by Mana Therapeutics for the treatment of acute myeloid leukemia (AML) and myelodysplastic syndromes (MDS), particularly in the relapsed or refractory post-transplant setting. The therapy consists of T cells specifically engineered to recognize tumor-associated antigens WT1, PRAME, and survivin. Upon administration, these T cells induce a targeted immune response leading to lysis of tumor cells expressing these antigens. The mechanism involves immunologic cytotoxicity through T lymphocyte replacement and activation against cancerous cells[1][3][4]. Clinical development included phase 1 trials for AML and MDS after allogeneic hematopoietic stem cell transplantation; however, development was discontinued due to manufacturing issues[2][3][4].
