Overview

Manfidokimab (AK120) is a humanized immunoglobulin G subclass 4 (IgG4) monoclonal antibody that targets the interleukin-4 receptor alpha subunit (IL-4Rα), a component shared by both the IL-4 and IL-13 receptors. By binding to IL-4Rα, it blocks signaling from both cytokines, thereby inhibiting the Th2 inflammatory pathway implicated in atopic dermatitis and other type 2 inflammatory diseases. Manfidokimab is being developed primarily for moderate-to-severe atopic dermatitis and has also been investigated for asthma and eosinophilic esophagitis. The drug has shown acceptable safety and preliminary efficacy in clinical trials[2][3][6].
