RVP-001 is a novel manganese-based magnetic resonance imaging (MRI) contrast agent developed as a gadolinium-free alternative to current general-purpose MRI contrast agents. It consists of the paramagnetic ion Mn2+ stabilized by the chelator PyC3A, enabling its use as an extracellular fluid agent for enhanced imaging during MRI procedures. The primary mechanism involves distribution in areas of expanded extracellular fluid volume, such as edema or fibrosis, producing increased signal on T1-weighted images. RVP-001 is being evaluated for safety and efficacy in detecting central nervous system (CNS) lesions—including brain tumors and multiple sclerosis—where it aims to provide equivalent or superior imaging performance compared to conventional gadolinium-based agents but with improved safety, especially for patients at risk of nephrogenic systemic fibrosis. Manganese is endogenously present and can be excreted via renal and hepatobiliary pathways, potentially offering advantages in patients with impaired kidney function[1][2][3][5][6][8].
