Overview

**Manganese-engineered Lactobacillus reuteri** is an experimental oral live biotherapeutic product for colorectal cancer. The product consists of *Lactobacillus reuteri* surface-engineered with manganese to enhance bacterial proliferation, glycerol metabolism, gastrointestinal survival, colorectal-tumor colonization, and production of antitumor metabolites, principally reuterin. In preclinical colorectal cancer models, MnLR was administered with glycerol as a metabolic substrate; this MnLR/Gly regimen promoted intratumoral bacterial and metabolite accumulation, selective tumor-cell killing, immunogenic cell death, dendritic-cell maturation, M1 macrophage polarization, and effector T-cell responses. The program remains preclinical; an enteric capsule formulation of MnLR plus glycerol, termed GlyMnLR, was evaluated in orthotopic rabbit colorectal cancer models.
