Mangifera indica L. extract (MIE), derived from the mango plant, is an experimental botanical therapeutic being investigated for the treatment of chronic immune-mediated conditions, specifically Inflammatory Bowel Disease (IBD). MIE functions as a multi-level immunomodulator by selectively antagonizing the pro-inflammatory Toll-like receptor (TLR) 2/4/6 axis and downregulating the adaptor protein MyD88. This action inhibits the nuclear translocation of NF-κB, leading to a significant reduction in the secretion of pro-inflammatory cytokines such as TNF-α and IL-6. Furthermore, MIE has been shown to impair monocyte recruitment by blocking their adhesion and transmigration across the activated endothelium and to promote a shift in macrophage polarization from the pro-inflammatory M1 phenotype toward a more homeostatic state.
