Overview

Mangiferin is a naturally occurring C-glucosyl xanthone (xanthonoid) primarily found in the leaves and bark of Mangifera indica (mango tree), as well as other plant species. It is a bioactive polyphenol with a wide range of pharmacological activities demonstrated in vitro and in vivo studies. These include antioxidative, anti-inflammatory, antitumor (anticancer), antibacterial, antiviral, immunomodulatory, antidiabetic (glucose-lowering), hepatoprotective (liver-protecting), cardioprotective and neuroprotective effects[1][4][7]. Mechanistically it acts through multiple molecular pathways such as reducing oxidative stress and inflammation; modulating AMPK/TBK1 signaling; restoring PDE3B stability to regulate lipid metabolism; inhibiting noncanonical NF-κB activation; and affecting glucose/lipid homeostasis[4]. Despite its promising therapeutic potential for chronic diseases like diabetes mellitus type 2/NAFLD/obesity/cardiovascular disease/neurodegenerative disorders/cancer/liver disease/kidney disease/skin disorders[1][4][7], clinical application has been limited by poor solubility and low oral bioavailability. Various formulations including nanoparticles or salt forms have been developed to improve these properties[4]. As of now there are no approved pharmaceutical drugs containing mangiferin but it is used in dietary supplements such as Zynamite.
