Overview

Manidipine is a dihydropyridine calcium channel blocker used primarily for the treatment of hypertension. It acts by selectively inhibiting L-type and T-type voltage-dependent calcium channels in vascular smooth muscle cells, leading to vasodilation and reduced blood pressure. Its selectivity for vascular tissue over cardiac tissue minimizes cardiac side effects at therapeutic doses. In addition to its antihypertensive effect, manidipine increases renal blood flow and promotes natriuresis (excretion of sodium), which may further contribute to its blood pressure-lowering action. The drug has a prolonged duration of action due to high receptor affinity and slow dissociation from binding sites[1][2][3][4].
