**mANK-101** is the murine surrogate of Ankyra Therapeutics' anchored IL-12 candidate ANK-101. It consists of a recombinant single-chain murine interleukin-12 fused to a phosphorylated alum-binding peptide and stably complexed with aluminum hydroxide. Following intratumoral injection, the complex forms a durable local depot that retains active IL-12 in the tumor microenvironment, limiting systemic exposure while stimulating IL-12 receptor signaling on T cells and natural killer cells. In mouse tumor models, mANK-101 induced IFN-gamma-driven antitumor immunity, tumor-infiltrating cytotoxic lymphocytes, myeloid-cell inflammatory remodeling, antigen-presentation programs, regression of injected tumors, and activity against noninjected lesions, particularly with checkpoint blockade or entinostat. It is a preclinical murine research surrogate rather than the human clinical candidate ANK-101.
