Mannan-MUC1 fusion protein is an experimental cancer immunotherapy comprised of the human tumor-associated antigen MUC1 covalently linked to the yeast cell-wall polysaccharide mannan. This fusion aims to enhance the immunogenicity of MUC1, which is overexpressed and abnormally glycosylated in many epithelial cancers, especially breast, colon, stomach, rectum, and ovarian cancers. Preclinical studies and early-phase trials have shown that mannan-MUC1 induces strong T cell-mediated and humoral immune responses by targeting antigen-presenting cells and stimulating cytotoxic T lymphocytes, with evidence of tumor stabilization in some patients. It has been tested both as a direct vaccine and in autologous dendritic cells pulsed with the fusion protein, with clinical trials showing robust cellular immune responses and a favorable safety profile[1][2][3][4][9].
