Overview

Mannosulfan is an experimental alkylating agent of the alkyl sulfonate class, structurally related to busulfan and treosulfan. It acts by forming interstrand and intrastrand cross-links in DNA, thereby inhibiting DNA replication and transcription, which leads to cell cycle arrest and apoptosis. Mannosulfan has been investigated primarily for its potential antineoplastic activity, especially in hematologic malignancies such as chronic myeloid leukemia (CML) and polycythemia vera. Research suggests it may be less toxic than busulfan. Its use has been mostly limited to Hungary, and it is not approved by the US FDA for cancer treatment[1][3][4][6].
