MAO-A siRNA is a small interfering RNA designed to silence the expression of Monoamine Oxidase-A (MAO-A), a mitochondrial enzyme involved in the degradation of monoamine neurotransmitters and the generation of reactive oxygen species (ROS). In preclinical research, specifically in models of heart failure with preserved ejection fraction (HFpEF), MAO-A siRNA has been used to demonstrate that MAO-A promotes cardiomyocyte senescence and diastolic dysfunction through the cGAS-STING signaling pathway. By reducing MAO-A levels, the siRNA attenuates mitochondrial ROS production, mtDNA leakage, and the subsequent activation of inflammatory and senescence-associated secretory phenotypes (SASP).
