Overview

MAP4343 is a synthetic neurosteroid, specifically the methyl ether derivative of pregnenolone (pregnenolone methyl ether, 3β-methoxy-pregnenolone). It acts as a stimulator of tubulin polymerization and neurite growth by binding to microtubule-associated protein 2 (MAP2), promoting the assembly of tubulin proteins into microtubules. Unlike traditional antidepressants, its mechanism does not involve direct modulation of neurotransmitter receptors but rather targets cytoskeletal dynamics in neurons. Preclinical studies have shown that MAP4343 has antidepressant and anxiolytic effects, improves memory deficits, and promotes recovery after spinal cord injury in animal models. It is being developed primarily for treatment-resistant depression and has also been investigated for alcohol use disorder and acute spinal cord injury. The drug cannot be converted into metabolites with hormonal activity, distinguishing it from other steroid derivatives[1][2][5].
