Overview

This is a combination therapy of **maplirpacept** (an engineered CD47 decoy receptor that blocks the "don't eat me" signal, thereby promoting phagocytosis by macrophages) and **rituximab** (a chimeric monoclonal antibody against CD20, widely used to target B-cells in hematologic cancers, especially non-Hodgkin lymphoma). Both drugs exert anticancer effects via distinct, non-overlapping immunological mechanisms; maplirpacept enhances macrophage-mediated clearance of tumor cells while rituximab promotes immune effector functions such as antibody-dependent cellular cytotoxicity. Together, they may act synergistically, and preclinical data suggest enhanced antitumor activity over single-agent therapy in diffuse large B-cell lymphoma (DLBCL) and possibly other hematological malignancies[1].
