**maplirpacept + pegylated liposomal doxorubicin** is a combination therapy consisting of maplirpacept, a recombinant fusion protein that acts as a CD47 inhibitor to block the "don't eat me" signal on cancer cells, and pegylated liposomal doxorubicin, a nanoparticle-encapsulated anthracycline chemotherapy. Maplirpacept (also known as ALX148) enhances the ability of the immune system’s phagocytes to engulf and destroy tumor cells by inhibiting the interaction between CD47 on cancer cells and signal-regulatory protein alpha (SIRPα) on macrophages, resulting in increased phagocytosis and anti-tumor immune responses. Pegylated liposomal doxorubicin delivers doxorubicin, a topoisomerase II inhibitor, via pegylated liposomes that circulate longer and preferentially deliver the drug to tumors, minimizing exposure to normal tissues and reducing cardiotoxicity. This combination is being investigated for cancer indications due to the potential synergistic effects of immune checkpoint blockade and cytotoxic chemotherapy[1][3][7].
