mAR-PSMA-PLGA is an experimental gene therapy candidate developed by researchers at Seoul National University for the treatment of prostate cancer, specifically targeting castration-resistant prostate cancer (CRPC) and enzalutamide-resistant cases. The therapeutic consists of a poly(lactic-co-glycolic acid) (PLGA) nanoparticle that is PEGylated and functionalized with a prostate-specific membrane antigen (PSMA) antibody to ensure targeted delivery to prostate cancer cells. The nanoparticle encapsulates a plasmid vector encoding a non-functioning mutant androgen receptor (mAR) with a 3-base pair deletion in the DNA-binding domain. Upon delivery, the constant overexpression of this non-functional AR competes with endogenous androgen receptor signaling, thereby suppressing tumor cell proliferation and reversing resistance to androgen deprivation therapies like enzalutamide.
