MAR001-HLE is a monoclonal antibody designed as a half-life-extended version of MAR001, targeting angiopoietin-related protein 4 (ANGPTL4). Developed by Marea Therapeutics, the drug incorporates specific Fc modifications to enhance its pharmacokinetic profile, potentially allowing for monthly low-volume dosing and reduced manufacturing costs. It is primarily being developed for the treatment of atherosclerotic cardiovascular disease (ASCVD) in patients with elevated triglycerides and remnant cholesterol. By inhibiting ANGPTL4, which is highly expressed in adipose tissue and acts as an inhibitor of lipoprotein lipase, MAR001-HLE aims to lower triglyceride levels and reduce cardiovascular risk. As of late 2025, the developer has aligned with the FDA on a Phase 1 PK bridging strategy to support advancement into Phase 3 development.
