Marchantia polymorpha ethanol extract is a plant-derived extract obtained by using ethanol as the solvent to isolate bioactive constituents from Marchantia polymorpha, a liverwort species long used in traditional medicine. The extract contains a range of phytochemicals, particularly phenolic compounds, tannins, flavonoids, and glycosides, which impart notable anti-inflammatory, antioxidant, antibacterial, and potential antitumor properties. In vitro studies show that the extract inhibits protein denaturation—a marker of anti-inflammatory activity—at levels comparable to standard drugs such as diclofenac sodium. Its antioxidant effects are demonstrated through dose-dependent free radical scavenging abilities in standard DPPH assays[1]. Data from in vivo and cellular models also demonstrate pro-apoptotic, anti-proliferative effects on hepatocellular carcinoma cells, with mechanisms involving reactive oxygen species (ROS) production, mitochondrial dysfunction, and caspase activation, suggesting its utility as a complementary anticancer agent[7]. The extract has further demonstrated moderate efficacy against both Gram-positive and Gram-negative bacteria. Its primary mechanism of action involves modulation of oxidative stress and downregulation of key inflammatory mediators like TNF-α, IL-1β, and IL-6, likely via inhibition of central pathways such as Jak/stat3 and influencing gene expression linked to antioxidant defense (e.g., upregulation of GPX1, TMX1, TXN)[4][1][7]. These actions are attributed mainly to a suite of bioactive phenolics, bis-bibenzyl derivatives (such as marchantin A), and related secondary metabolites.
