Drug intelligence / Profile preview
Maresin 2 (MaR2; 13R,14S-dihydroxy-docosahexaenoic acid) is an endogenous specialized pro-resolving mediator (SPM) biosynthesized from the omega-3 polyunsaturated fatty acid docosahexaenoic acid (DHA) by macrophages. Its biosynthesis is mediated by 12-lipoxygenase (12-LOX) and soluble epoxide hydrolase (sEH). Maresin 2 possesses potent anti-inflammatory, pro-resolving, and tissue-regenerative properties. It acts as an agonist of the leukotriene B4 receptor 1 (LTB4R/BLT1) and regulates downstream pathways including the upregulation of TREM2 in macrophages, suppression of the NLRP3 inflammasome, and activation of cardiac autophagy via ULK1 O-GlcNAcylation. Preclinical studies have demonstrated its therapeutic potential in mitigating cardiac inflammation and remodeling in diabetic cardiomyopathy, heart failure with preserved ejection fraction (HFpEF), and myocardial infarction, as well as promoting mucosal wound healing in colitis and alleviating inflammatory and neuropathic pain.
