MART-1 + gp100 + tyrosinase is a multi-antigen cancer vaccine combination primarily investigated for the treatment of metastatic melanoma. It consists of specific immunogenic peptides derived from three well-characterized melanoma-associated antigens (MAAs): Melanoma Antigen Recognized by T cells 1 (MART-1), Glycoprotein 100 (gp100), and Tyrosinase. The vaccine is typically administered with an adjuvant, such as Montanide ISA-51, to enhance the immune response. Its mechanism of action involves the induction of a robust, antigen-specific cytotoxic T-lymphocyte (CTL) response. By presenting these peptides in the context of Human Leukocyte Antigen (HLA) molecules (most commonly HLA-A2), the vaccine primes the immune system to recognize and destroy melanoma cells that overexpress these proteins. Development has been largely driven by the National Cancer Institute (NCI) through various Phase 1 and Phase 2 clinical trials.
