The MART-1 + gp100 + tyrosinase + dendritic cell vaccine is an autologous cellular immunotherapy designed for the treatment of advanced or metastatic melanoma. The vaccine is produced by harvesting a patient's peripheral blood mononuclear cells (PBMCs), differentiating them ex vivo into mature dendritic cells (DCs), and then pulsing these DCs with MHC class I-restricted peptides derived from three melanocyte-lineage antigens: MART-1 (Melanoma-associated antigen recognized by T cells 1), gp100 (Melanocyte protein PMEL), and tyrosinase. Upon re-administration, the matured dendritic cells function as professional antigen-presenting cells that migrate to lymph nodes and prime naive CD8+ T lymphocytes. This process induces the expansion of tumor-specific cytotoxic T cells capable of identifying and eliminating melanoma cells that express these specific antigens. By targeting multiple antigens simultaneously, the vaccine aims to minimize the risk of tumor immune escape resulting from antigen loss or heterogeneity.
