MART-1 peptide-pulsed dendritic cell vaccine is an autologous cellular immunotherapy developed by M.D. Anderson Cancer Center for the treatment of metastatic melanoma. The therapy involves harvesting a patient's own dendritic cells and pulsing them with MART-1 (Melanoma Antigen Recognized by T cells 1) peptides to enhance their ability to present tumor antigens. These matured dendritic cells are administered intravenously to stimulate and sustain the anti-tumor activity of adoptively transferred tumor-infiltrating lymphocytes (TILs). This approach is typically used in a regimen following lymphodepletion with cyclophosphamide and fludarabine, and is accompanied by high-dose interleukin-2 (IL-2) to promote T-cell expansion and persistence in vivo. The treatment is specifically designed to enhance the immune system's ability to recognize and destroy melanoma cells by providing potent antigen presentation.
