This combination therapy consists of autologous dendritic cells (DCs) that have been pulsed with HLA-A*0201-restricted peptides derived from two well-characterized melanoma-associated antigens: MART-1 (Melanoma-associated antigen recognized by T cells 1) and gp100 (Glycoprotein 100). Dendritic cells are professional antigen-presenting cells (APCs) harvested from the patient via leukapheresis, matured ex vivo, and loaded with these specific tumor-associated antigens. Upon re-administration into the patient, these pulsed DCs present the MART-1 and gp100 epitopes to the host's immune system, specifically priming and expanding CD8+ cytotoxic T lymphocytes (CTLs). This targeted activation is designed to induce a specific and robust anti-tumor immune response against melanoma cells expressing these antigens. The therapy was primarily investigated by the National Cancer Institute (NCI) for patients with advanced or metastatic melanoma, often in combination with other immunomodulators like interleukin-2.
