Overview

MART-10 (19-nor-2α-(3-hydroxypropyl)-1α,25-dihydroxyvitamin D3) is a synthetic, less-calcemic analog of 1α,25-dihydroxyvitamin D3 (calcitriol), the active form of vitamin D. Developed by researchers at Teikyo University and Boston University, MART-10 acts as an agonist of the vitamin D receptor (VDR). It has demonstrated significantly higher potency than calcitriol in preclinical studies across various cancer types—including breast, head and neck, pancreatic, liver, and anaplastic thyroid cancers—by inducing cell cycle arrest and apoptosis and inhibiting metastasis, while causing substantially less hypercalcemia. Additionally, MART-10 is being evaluated in a Phase 2 clinical trial sponsored by Pediatric Endocrine Clinics for the secondary prevention of type 1 diabetes, prediabetes, celiac disease autoimmunity, and thyroid autoimmunity in at-risk children and adolescents, leveraging its immunomodulatory properties.
