MAS825 is a bispecific human IgG1 monoclonal antibody that targets and inhibits both interleukin-1 beta (IL-1β) and interleukin-18 (IL-18), two key pro-inflammatory cytokines involved in the pathogenesis of various autoinflammatory, autoimmune, and inflammatory diseases. By simultaneously blocking IL-1β and IL-18, MAS825 aims to reduce inflammation more effectively than single-cytokine inhibitors. It is being developed primarily for rare autoinflammatory conditions such as systemic juvenile idiopathic arthritis (sJIA) complicated by macrophage activation syndrome (MAS) and interstitial lung disease, as well as for other disorders including NLRC4-Gain of Function syndrome, XIAP deficiency, CDC42 mutations, COVID-19 pneumonia, lymphoproliferative disorders, hidradenitis suppurativa, enterocolitis associated with infantile autoinflammation with enterocolitis (AIFEC), and atherosclerotic cardiovascular disease in patients with clonal hematopoiesis of indeterminate potential (CHIP). Clinical studies have shown promising results in drug-resistant sJIA/MAS cases with high serum IL-18 levels[2][4][5][7].
