MASTL RNAi refers to the use of RNA interference (RNAi) to silence the expression of Microtubule Associated Serine/Threonine Kinase Like (MASTL), also known as Greatwall kinase. In the context of Acute Myeloid Leukemia (AML), MASTL has been identified as a potential therapeutic target due to its significantly higher expression in AML patients compared to healthy individuals. Suppression of MASTL via RNAi has been shown to reduce cell viability and colony formation in AML cell lines such as THP-1 and OCI-AML3. In vivo studies using xenograft mouse models demonstrate that MASTL knockdown significantly reduces tumor growth and prolongs survival. Mechanistically, the silencing of MASTL induces the downregulation of Survivin and the upregulation of Puma, while also suppressing the expression of S100A8 and S100A9, proteins associated with resistance to venetoclax. Consequently, MASTL RNAi is being explored as a strategy to both treat AML and overcome resistance to venetoclax-based therapies.
