Drug intelligence / Profile preview
The matching vehicle is a topical ointment formulation developed by Timber Pharmaceuticals to serve as the placebo control in clinical trials for TMB-001 (topical isotretinoin). It is chemically identical to the TMB-001 ointment but lacks the active pharmaceutical ingredient, isotretinoin. In the Phase 3 ASCEND trial for congenital ichthyosis—specifically autosomal recessive congenital ichthyosis (ARCI) and X-linked recessive ichthyosis (XLRI)—this vehicle demonstrated an unexpectedly high therapeutic response. This high vehicle response contributed to the trial failing to meet its primary and key secondary endpoints, as the difference between the active treatment and the placebo was not statistically significant. The vehicle's composition is designed to mimic the emollient properties of the active treatment without the retinoid-mediated effects.
