Matrix M is a saponin-based vaccine adjuvant developed by Isconova and further developed by Novavax. It consists of two distinct fractions of saponins purified from the Quillaja saponaria Molina tree, combined with cholesterol and phospholipids to form 40-nm open cage-like nanoparticles. This adjuvant technology is designed to enhance and modulate the immunogenicity of antigens, leading to a stronger, longer-lasting, and broader immune response. It functions by inducing early activation of innate immune cells at the injection site and in draining lymph nodes, facilitating antigen uptake and presentation to T cells, and promoting a Th1-dominant immune response. Matrix M is a critical component in various vaccine candidates for infectious diseases, including those targeting rabies, malaria, influenza, and COVID-19.
