Matrix-M2 adjuvant is a saponin-based vaccine adjuvant developed by Novavax. It consists of 40-nm open cage-like nanoparticles made from saponins purified from the Quillaja saponaria Molina tree (Chilean soap bark tree), combined with cholesterol and phospholipids. The adjuvant enhances immune system responses to vaccines by inducing high levels of broadly neutralizing antibodies and enhancing both B-cell and T-cell immunity. Matrix-M2 works by creating early activation of innate immune cells at the injection site and in the draining lymph nodes, which translates into improved magnitude and quality of antibody responses, broadened epitope recognition, and induction of a Th1-dominant immune response. It has antigen dose-sparing properties, allowing for the use of less antigen per vaccine dose, and has been shown to have an acceptable safety profile in clinical trials.
