The mature non-polarized dendritic cell vaccine is an autologous cellular immunotherapy developed by Pawel Kalinski and his team, primarily evaluated at the University of Pittsburgh. This vaccine consists of dendritic cells (DCs) derived from a patient's own monocytes, which are matured using a standard cytokine cocktail (typically including IL-1β, TNF-α, IL-6, and PGE2) that does not induce the high-level IL-12p70 production characteristic of "polarized" Alpha-type-1 DCs. These mature, non-polarized cells are loaded with melanoma-associated tumor peptides and proteins. When administered intralymphatically, the vaccine is designed to migrate to the lymph nodes and present these antigens to naive T cells, thereby stimulating a systemic cytotoxic T lymphocyte (CTL) response against metastatic melanoma cells. It was notably used as a comparator arm in a Phase I clinical trial (NCT00053391) to assess the relative safety and immunogenicity of different dendritic cell maturation states.
