Overview

Matuzumab is a humanized monoclonal antibody developed for the treatment of various cancers. It specifically targets the epidermal growth factor receptor (EGFR), binding with high affinity and competitively inhibiting ligand binding. By blocking EGFR, matuzumab interferes with downstream signaling pathways that promote tumor cell proliferation, survival, and angiogenesis. The drug was investigated in multiple solid tumors including gastric cancer, non-small cell lung cancer, ovarian cancer, cervical cancer, colorectal cancer, head and neck cancer, and esophageal adenocarcinoma. Despite being well tolerated in clinical trials—most commonly causing skin toxicities such as rash and acne—matuzumab did not demonstrate significant clinical activity as monotherapy or in combination regimens compared to standard therapies. Development was discontinued after phase II trials due to lack of efficacy[1][3][6][8][9].
