A fixed combination of **mavorixafor**, a selective, orally available CXC chemokine receptor 4 (CXCR4) antagonist, and **axitinib**, an oral vascular endothelial growth factor receptor (VEGFR) tyrosine kinase inhibitor (TKI), evaluated in combination therapy for advanced clear cell renal cell carcinoma (ccRCC). **Mavorixafor** blocks CXCR4-mediated signaling, increasing mobilization of neutrophils and lymphocytes, and is FDA-approved for WHIM syndrome. **Axitinib** inhibits angiogenesis by selectively targeting VEGF receptors. The combination is designed to overcome resistance and improve efficacy relative to VEGFR TKI monotherapy in ccRCC; clinical evidence from phase I/II studies indicates improved progression-free survival and objective response rates with manageable safety compared to axitinib alone.
