This is a combination therapy consisting of **mavorixafor (X4P-001)**, an orally bioavailable, selective, allosteric inhibitor of the chemokine receptor **CXCR4**, and **nivolumab**, a PD-1-targeting immune checkpoint inhibitor monoclonal antibody. Mavorixafor blocks the CXCR4/CXCL12 axis, disrupting recruitment of immunosuppressive cells and increasing cytotoxic T cell infiltration in the tumor microenvironment. Nivolumab blocks PD-1 on T cells, enhancing antitumor immunity by preventing tumor-induced T cell inactivation. This combination is being investigated primarily for advanced clear cell renal cell carcinoma (ccRCC) in patients who did not respond to nivolumab monotherapy, with evidence of immune modulation and tumor immune infiltration.
