Overview

Mavrostobart + gemcitabine is a combination therapy consisting of mavrostobart (PT199), a humanized monoclonal antibody targeting the ecto-5'-nucleotidase (CD73) enzyme, and gemcitabine, a nucleoside analog chemotherapy. Mavrostobart inhibits the conversion of adenosine monophosphate (AMP) to immunosuppressive adenosine in the tumor microenvironment, thereby enhancing anti-tumor immune responses and potentially overcoming resistance to checkpoint inhibitors. Gemcitabine acts as a cytotoxic agent by inhibiting DNA synthesis through incorporation into DNA and inhibition of ribonucleotide reductase. This combination is being developed by Phanes Therapeutics and is currently being evaluated in the Phase 1/2 MORNINGSTAR clinical trial for the treatment of advanced solid tumors, including pancreatic ductal adenocarcinoma (PDAC) and non-small cell lung cancer (NSCLC), often in combination with nab-paclitaxel.
