**MAVU-104** is a first-in-class, orally active small molecule inhibitor of ectonucleotide pyrophosphatase/phosphodiesterase 1 (**ENPP1**), an enzyme that negatively regulates the stimulator of interferon genes (**STING**) pathway by hydrolyzing the second messenger 2'3'-cyclic GMP-AMP (cGAMP). By inhibiting ENPP1, MAVU-104 enables highly controlled enhancement of STING signaling in tumors and lymph nodes, promoting innate immune activation, type I interferon production, and antitumor immune responses without the need for intratumoral injections. Originally developed by Mavupharma, it was acquired by **AbbVie** in 2019 as part of their early-stage oncology pipeline to advance immuno-oncology therapies, with preclinical data demonstrating tumor shrinkage and synergy with other immunotherapies in mouse cancer models.[1][3][4]
