MAX-4 is a small molecule multi-kinase inhibitor being developed by Guangzhou Zaiji Medical Technology (Maxinovel) for the treatment of acute myeloid leukemia (AML). The drug primarily targets Fms-like tyrosine kinase 3 (FLT3) and fibroblast growth factor receptors (FGFR), and it also exhibits inhibitory activity against hematopoietic progenitor kinase 1 (HPK1). By simultaneously inhibiting these pathways, MAX-4 is designed to address resistance mechanisms in AML, particularly those mediated by FGFR signaling or FLT3 mutations. The compound, also identified by the code MAX-40279, has received Orphan Drug Designation from the U.S. FDA for AML and is being evaluated both as a monotherapy and in combination with other agents, such as the bispecific antibody KN046. It is currently in Phase 2/3 clinical development for hematological malignancies.
