Overview

MAXY-G34 is a PEGylated recombinant human granulocyte colony-stimulating factor (G-CSF) variant developed by Maxygen for the treatment of chemotherapy-induced neutropenia. The drug was engineered using directed evolution to include five specific amino acid substitutions (at positions 16, 34, 40, 105, and 159) that introduce non-naturally occurring lysine residues, serving as sites for site-specific PEGylation with 5 kDa monomethoxy-polyethylene glycol (mPEG) groups. MAXY-G34 acts as an agonist of the Colony Stimulating Factor 3 Receptor (CSF-3R), stimulating the bone marrow to produce and release neutrophils. Although it demonstrated positive results in Phase IIa clinical trials for breast cancer patients, showing good safety and tolerability, its development has been discontinued. It was also investigated as a potential medical countermeasure for Hematopoietic Acute Radiation Syndrome (H-ARS).
