Mazdutide is a long-acting, once-weekly injectable dual agonist of the glucagon-like peptide 1 receptor (GLP-1R) and the glucagon receptor (GCGR), developed primarily for chronic weight management and type 2 diabetes. It is an analog of oxyntomodulin with a fatty acid side chain to prolong its action. Mazdutide works by activating both GLP-1R and GCGR, leading to reduced appetite, delayed gastric emptying, increased energy expenditure, enhanced fatty acid oxidation and lipolysis, improved glucose control, and reduction in liver fat content. Clinical trials have demonstrated robust weight loss effects as well as improvements in multiple cardiometabolic risk factors. The drug has been well tolerated in studies with gastrointestinal side effects being most common[1][2][4][5][7].
