Overview

MB-204 is a novel, fluorinated small molecule derivative of Istradefylline, an FDA-approved adenosine A2a receptor antagonist. Developed by Marvel Biosciences, MB-204 is designed to maximize therapeutic benefits by acting faster and more effectively at lower doses than existing treatments. Its primary mechanism involves antagonism of the adenosine A2a receptor—a target implicated in numerous neurological and psychiatric disorders. Preclinical studies have demonstrated that MB-204 can reverse symptoms and underlying disease mechanisms in models of Alzheimer’s disease, depression, anxiety, autism spectrum disorder (ASD), Rett syndrome, Fragile X syndrome, and attention deficit hyperactivity disorder (ADHD). The compound has shown durable post-treatment effects in animal models for Rett syndrome and autism. As of mid-2025, MB-204 is advancing toward Phase 1 clinical trials with ongoing preclinical research for several indications[1][3][4][5][6][8].
