Overview

MB-dNPM1-TCR.1 is an autologous, ex vivo expanded T cell receptor-engineered (TCR) T cell therapy developed by Miltenyi Biomedicine for the treatment of relapsed or refractory acute myeloid leukemia (AML) with mutated nucleophosmin (NPM1). The therapy involves collecting a patient's own T cells and genetically modifying them using a lentiviral vector to express a specific T cell receptor that recognizes dNPM1 peptides presented by HLA-A*02:01 on leukemic myeloid cells. These engineered cells are then expanded and infused back into the patient after lymphodepleting chemotherapy, where they target and eliminate malignant AML cells expressing the NPM1 mutation[3][7][6]. The product is currently in Phase I/II clinical trials as second-line or greater therapy for AML[2][3][4].
