MB07133 + PD-1 is an investigational combination therapy consisting of MB07133, a liver-targeted cytarabine mononucleotide (araCMP) prodrug, and sintilimab, a programmed cell death protein 1 (PD-1) immune checkpoint inhibitor. Developed by Xintong Pharmacy, this combination is being evaluated for the treatment of primary liver cancer, specifically advanced hepatocellular carcinoma (HCC). MB07133 utilizes the HepDirect technology platform to mask the negative charges of the monophosphate group, allowing it to selectively enter hepatocytes where it is cleaved by cytochrome P450 3A (CYP3A) enzymes. Once activated, the resulting araCMP is phosphorylated by intracellular kinases to cytarabine triphosphate (araCTP), which inhibits DNA polymerase and induces DNA damage in proliferating tumor cells. Sintilimab blocks the interaction between PD-1 and its ligands (PD-L1 and PD-L2), thereby restoring T-cell-mediated anti-tumor immune responses. The combination is currently in Phase 1/2a clinical development to assess its safety, tolerability, and efficacy.
