Overview

MBC-11 is a first-in-class bone-targeting therapeutic conjugate developed by Osteros Biomedica (licensed from MBC Pharma) for the treatment of cancer-induced bone disease (CIBD) and bone metastases. The molecule consists of the bisphosphonate etidronate covalently linked to the antimetabolite cytarabine (ara-C). This design utilizes the high affinity of etidronate for bone hydroxyapatite to concentrate the cytotoxic cytarabine at sites of active bone resorption and tumor growth. By targeting the bone microenvironment, MBC-11 aims to provide potent anti-tumor activity while reducing systemic exposure and associated toxicities. A Phase 1 dose-escalation study (NCT02673060) demonstrated that the drug was well-tolerated, with myelosuppression as the primary toxicity, and showed promising reductions in the metabolic activity of bone-associated cancer cells.
